fa بررسی هم‌افزایی نانوژل‌های کیتوزان لود شده با وانکومایسین و اینترفرون گاما بر بیان ژن TNF-α در موش‌های آلوده شده با استافیلوکوک اورئوس مقاوم به متی‌سیلین (MRSA) Synergistic effect of vancomycin and interferon-gamma-loaded chitosan nanogels on TNF-α gene expression in methicillin-resistant Staphylococcus aureus (MRSA) -infected mice میکروبیولوژی مواد غذایی پژوهشي Research <div style="text-align: justify;">مشکل فزاینده مقاومت آنتی&rlm; بیوتیکی استافیلوکوکوس اورئوس، تلاش‌های تازه‌ای را برای کشف عوامل ضدمیکروبی جدید برانگیخته است. یکی از این روش‌های نوین، استفاده از نانوذرات است. هدف از این مطالعه، سنتز نانوژل&rlm;های کیتوزان لودشده با وانکومایسین و اینترفرون گاما (γ(IFN- و بررسی بیان ژن‌ TNF-α در موش&rlm;های آلوده به استافیلوکوک اورئوس مقاوم به متی&rlm;سیلین (MRSA) می‌باشد. پس از سنتز نانوژل&rlm; کیتوزان و تایید آن، بارگذاری وانکومایسین و γIFN- در این نانوژل&rlm;ها انجام و توسط آنالیز FTIR تایید شد. سپس، فعالیت ضدباکتریایی نانوژل&rlm;ها ارزیابی شد. حداقل غلظت مهارکنندگی (MIC) در مورد نانوژل لود شده با وانکومایسین در pHهای 6/5 و 7/4، به ترتیب µg/ml 8 و 64 به‌دست آمد. MIC در مورد نانوژل کیتوزان و نانوژل لود شده با γIFN-، 1024 و در مورد وانکومایسین آزاد، µg/ml 1 در هر دو pH محاسبه شد. سپس، 48 راس موش&rlm; به 6 گروه کنترل، MRSA، نانوژل، نانوژل دارو، نانوژل γIFN- و نانوژل&rlm;های (دارو و γIFN-) تقسیم شدند. پس از آلوده‌شدن گروه&rlm;ها (غیر از کنترل) با MRSA، mg/kg100 از نانوژل، نانوژل دارو، نانوژل γIFN- و نانوژل&rlm;های (دارو و γIFN-)، به ترتیب به گروه&rlm;های 3 تا 6 اضافه شد. در ادامه، در ساعات 6، 12 و 24، از هر گروه تعدادی موش بیهوش شدند و طحال آن&rlm;ها جهت بررسی بیان ژن TNF-α  به روش RT-PCR ارزیابی شد. بر طبق نتایج، در گروه&rlm;های 4، 5 و 6 در مقایسه با گروه MRSA، بیان ژن&rlm; TNF-α پس از 24 ساعت کاهش معنی&rlm;دار نشان می&rlm;دهد. بنابراین، می&rlm;توان نتیجه گرفت تزریق همزمان نانوژل دارو و نانوژل γIFN- در موش‌های آلوده تاثیر بسیار بیشتری در مقایسه با تزریق تکی هر کدام از این نانوژل&rlm;ها دارد.</div> <div style="text-align: justify;">Increasing antibiotic resistance of Staphylococcus aureus has led to attempts to discover new antimicrobial agents, including the use of nanoparticles. The aim of this study was to synthesize vanosomycin and interferon gamma (IFN-γ) loaded chitosan nanogels and to evaluate the expression of TNF-α gene in mice infected with methicillin-resistant Staphylococcus aureus (MRSA). After synthesis and confirmation of chitosan nanogels, loading of vancomycin and IFN-γ in these nanogels was performed and confirmed by FTIR analysis. Then the antibacterial activity of nanogels was evaluated. At pHs of 6.5 and 7.4, the minimum inhibitory concentration (MIC) for vancomycin-loaded nanogels was 8 and 64 µg/ml, respectively. The MIC was calculated to be 1024 µg/ml for chitosan nanogels and IFN-γ-loaded nanogels and 1 µg/ml for free vancomycin. Then 48 mice were divided into 6 groups: control, MRSA, nanogels, drug nanogels, IFN-γ nanogels and nanogels (drug and IFN-γ). After infecting the groups (except control) with MRSA, 100 mg/kg of nanogels, drug nanogels, IFN-γ nanogels and nanogels (drug and IFN-γ) were added to groups 3 to 6, respectively. Finally, at 6, 12 and 24 hours, a number of mice from each group were anesthetized and their spleen was evaluated for TNF-α gene expression by RT-PCR. The results showed that in groups 4, 5 and 6, compared to the MRSA group, TNF-α gene expression showed a significant decrease after 24 hours.
